Ok so I've got a pumpkin that has certain characteristics that I would like to keep going. I guess my question is in order to do this do I need to isolate the pumpkin plant next year to keep it true or just try hand pollination? Thanks

You will only need to control the pollination, But and this is probably more important
The fruit you have now. Was the pollination controlled, if you didn't control it then you won't know the male side and therefore those characteristics could show up.

Then you will need to grow a good sized number of seeds. Hand pollinate each with a male flower from each. Select the one with closest to characteristics you want. Do the same for the next year.
