Should I prune secondary or tertiary vine coming from stem of a good fruit? How soon?

It should be trimmed off by "basketball" size. Shortly thereafter, the root beneath the fruit will have to be cut to allow the stem to rise with the growing pumpkin.

I've always let mine continue growing until I need to remove it because it is in the pumpkin's way or putting a stress on the pumpkin stem. If this is wrong or not as good could someone let me know why.
