No, don't use manure/humus! Manure can contain bad bacteria.

Manure can contain bad bacteria, but as long as your dissolved oxygen levels are maintained above 6 mg/l for the entire brew cycle then you should be fine. You're also spraying on to your pumpkin, which you don't plan on eating, so certain facultative anaerobes like e. coli or salmonella aren't as much of an issue.

All that being said, if you have a good vermicompost source or other compost source, then you're probably better off using that instead.
